Glucose-6-Phosphate Dehydrogenase of Trypanosomatids: Characterization, Target Validation, and Drug Discovery

G6PDH is important for defense against oxidative stress in bloodstream form T. brucei. Wild-type (WT) trypanosomes and cells in which partial depletion of G6PDH was induced by RNAi were grown in regular HMI-9 medium. After growth for different periods of time as indicated, cells were collected by centrifugation and resuspended in non-reducing medium (i.e., without β-mercaptoethanol and cysteine) and the cell density was determined. Cell suspensions were then incubated for 1.5 h in the presence of different concentrations of H2O2 after which the cell density was determined again.
