Heme-induced HO-1 expression is NADPH dependent. Rat AMs (1 × 106/mL) were pretreated with DPI (10 μM) followed by incubation in the absence or presence of heme (10 μM) for 24 h. Incubations were terminated by the addition of lysis buffer and lysates were subjected to immunoblotting with anti-HO-1 and anti-actin antibodies. Results are demonstrated as the HO-1/actin ratio. The data shown are representative of three independent experiments. The mean ± S.D. is presented. when compared with controls. when compared to cells treated with heme.
