Celecoxib Improves Host Defense through Prostaglandin Inhibition during Histoplasma capsulatum Infection

Celecoxib treatment modulates endogenous PGE2 and LTB4 production in the lungs of H. capsulatum-infected mice. Lipid mediators in the lung parenchyma were measured using enzyme immunoassay (a) PGE2, (b) LTB4. The lungs of mice infected i.t. with sublethal H. capsulatum inoculum and daily p.o. treatment with water or celecoxib (1 mg/kg/0.5 mL) were removed at 2, 14, or 28 days after infection and homogenized in medium in the presence of indomethacin. A group of noninfected mice receiving water (solid line) and noninfected mice treated with celecoxib (dotted line) were used as controls. The data are presented as the means ± SEM representative of three independent experiments . One-way ANOVA (Tukey's multiple comparison test) was used. *PBS + H2O versus Hc + H2O or versus Hc + celecoxib; #Hc + H2O versus Hc + celecoxib .
