Anti-Inflammatory Mechanism of Polyunsaturated Fatty Acids in Helicobacter pylori-Infected Gastric Epithelial Cells

Figure 3

Effects of PUFAs on H. pylori-induced activation of NF-κB and AP-1. (a) AGS cells were infected with H. pylori for indicated time periods (0.5, 1, 2, and 4 h). Nuclear extracts were harvested and the DNA-binding activities of NF-κB- and AP-1 were determined by electrophoretic mobility shift assay. (b) AGS cells were pretreated with individual fatty acid (100  M) for 24 h and stimulated with H. pylori for 1 h. (c) H. pylori-induced degradation of IκBα was analyzed by western blotting at the indicated time points (15, 30, 45, 60, and 120 min; upper panel). AGS cells were pretreated with individual fatty acid (100  M) for 24 h and infected with H. pylori for 30 min (lower panel). Actin was used as a loading control.
