Effect of Th17 polarizing conditions on the cytokine secretion of CD4⁺ T cells. Human CD4⁺ T cells were cultured with anti-CD3/anti-CD28 with or without Th17 polarizing conditions for five days. The concentration of the indicated cytokines in the cell culture supernatant was measured by CBA (IL-17A, IL-9, IL-8, IFN-γ) and ELISA (IL-22). The absolute concentration [pg/mL] of IL-17A, IL-22, IL-9, IL-8, and IFN-γ in the supernatant of CD4⁺ T cells cultured with anti-CD3/anti-CD28 (○) or in addition of Th17 polarizing conditions (●) during the time course is shown as mean ± SEM. *represents significance, 𝑃<0.05, Mann-Whitney 𝑈 test, and 𝑛=15.
