IL-1β and TNFα Promote Monocyte Viability through the Induction of GM-CSF Expression by Rheumatoid Arthritis Synovial Fibroblasts

Neutralisation of GM-CSF but not M-CSF nor IL-34 reduces monocyte viability induced by IL-1β and TNFα prestimulated SF conditioned media. CD14+ cells were cultured 3 days in presence of M-CSF (25 ng/mL), IL-34 (50 ng/mL), GM-CSF (10 ng/mL), or conditioned media from SF prestimulated or not (NS) with TNFα or IL-1β for 24 hours. Blocking antibodies against M-CSF, IL-34, and GM-CSF, at 2, 10, and 5 μg/mL, respectively, were added at the same time. Results are given as the mean percentage of viability (± SEM) considering M-CSF as control (100%) ( = 3–5 patients). ; ; .
