ATP-Binding Pocket-Targeted Suppression of Src and Syk by Luteolin Contributes to Its Anti-Inflammatory Action

The effects of luteolin on the pathway upstream of NF-κB activation. ((a), (b), and (c)) RAW264.7 cells (5 × 106 cells/mL) were incubated with LPS (1 μg/mL) in the presence or absence of luteolin for the indicated times. After preparing whole lysates, the levels of total or phosphorylated IκBα, Src, Syk, ERK, p38, and JNK were determined by immunoblotting. (d) The inhibitory effects of luteolin on Src and Syk activity were determined using a conventional kinase assay with purified Src and Syk. All data are expressed as the mean ± SD of experiments. and compared to the control group.
