Alterations in Nitric Oxide Synthase in the Aged CNS

NADPH-d-positive neurons in the rat inferior colliculus. The number of NADPH-d-positive neurons in the inferior colliculus was significantly higher in aged than in younger rats. NADPH-d-positive neurons are dark purple. Frozen sections of 40 μm thickness were made in the coronal plane. The histochemical detection for NADPH-d activity was performed as follows: after free-floating sections were incubated for 10 min at room temperature in 0.05 M Tris buffer (pH 8.0), sections were incubated for 60 min at 37°C in the 0.05 M Tris buffer containing 0.3% (v/v) Triton X-100, 0.1 mg/mL nitroblue tetrazolium, and 1.0 mg/mL -NADPH.
