Curcumin Pretreatment Induces Nrf2 and an Antioxidant Response and Prevents Hemin-Induced Toxicity in Primary Cultures of Cerebellar Granule Neurons of Rats

Figure 6

The protective effect of curcumin on hemin-induced cell death was prevented by the enzyme inhibitors tin mesoporphyrin (SnMP) or buthionine sulfoximine (BSO). CGNs were incubated with 10 and 15 μM curcumin for 24 h. The culture medium was removed and the inhibitors (a) SnMP or (b) BSO were added (15 min and 1 h, resp.), before the addition of 30 μM hemin. Subsequently these compounds were replaced by fresh medium and the incubation was continued in presence of inhibitors for 24 h. Finally, cell viability was quantified by MTT reduction. Data are expressed as mean ± SEM, . versus control (untreated), versus hemin.
