miR-34a targets SIRT1. (a) The expression of SIRT1 in HG/PA-treated INS-1 cells was assessed by western blot analysis. Representative bands of SIRT1 are shown. Histogram represents SIRT1 expression normalized to the corresponding tubulin. (b) Western blot analysis revealed that inhibition of miR-34a increased the SIRT1 expression. (c) INS-1 cells were stained with Annexin V-FITC and PI and then subjected to flow cytometry to measure apoptosis. The apoptotic rate was calculated as the percentage of Annexin V-positive cells divided by the total number of cells. (d) The generation of intracellular ROS was assessed using DCFH-DA.
