Conditional Expression of Human PPARδ and a Dominant Negative Variant of hPPARδ In Vivo

Mice conditionally express hPPARδ or hPPARδΔAF2 transgenes in response to feeding I3C. Animals ( per group, 10 weeks old, mixed male and female) were placed either on control diet or a diet containing I3C (0.25% (w/w)) for 5 days. (a) hPPARδ and hPPARδΔAF2 exhibit low basal mRNA expression (open bars) and highly inducible I3C-dependent expression (black bars) in mouse liver. (b) hPPARδ and hPPARδΔAF2 protein is expressed in livers in response to dietary supplementation with I3C. (c) hPPARδ (hδ) and hPPARδΔAF2 (DN) mRNA expression in various mouse organs in response to normal diet (open bars) and I3C-supplemented diet (black bars). Statistical significance where indicated was analysed using Mann-Whitney nonparametric test in GraphPad Prism version 5.0c, (GraphPad Software, San Diego, CA, USA).
