Figure 8: High-glucose-induced DNA binding of DIG-labeled NFκB. NFκB-DNA binding activity was significantly increased with high glucose. Pioglitazone significantly reduces high-glucose-induced NFκB binding activity. Gefitinib has a similar effect. HK2 cells were incubated for 72 hrs with 5 mM glucose media (Control), high glucose (HG) ± Piog (10μM) ± gefitinib (0.5μM). Nuclear extract preparation and EMSA are as described in Section 2.
