15-Deoxy-Δ12,14-Prostaglandin J2 Inhibits Homing of Bone Marrow-Derived Mesenchymal Stem Cells Triggered by Chronic Liver Injury via Redox Pathway

Figure 4

The suppressive effect of 15d-PGJ2 on BMSCs migration occurs via ROS production. (a) Serum-starved BMSCs were incubated with 5 μM 15d-PGJ2 with or without 2.5 mM NAC (inhibitor of ROS) for 1 hour before treatment and then determined the effect of 15d-PGJ2 on BMSCs migration. ((b) and (c)) BMSCs were loaded with DCFH-DA and further treated with 15d-PGJ2 at different dose levels and durations. Representative images of fluorescence for ROS (green) in BMSCs (c). Fluorescence was measured using high content analysis (b). , compared with control. , compared with 15d-PGJ2 group without NAC (). Scale bars, 50 μm.
