The TGF-β signaling pathway. The TGF-β signaling pathway is activated by binding of TGF-β to a type II receptor, which facilitates the recruitment and phosphorylation of the type I receptor. This pTGF-βRI activates either SMAD2 or SMAD3 by phosphorylation. The phosphorylated SMAD2/3 forms a complex with SMAD4 and transports to the nucleus where it influences SMAD target gene transcription. SMAD7 can inhibit TGF-β signaling through inhibition of TGF-βRI phosphorylation.
