Lentiviral-Mediated shRNA Silencing of PDE4D Gene Inhibits Platelet-Derived Growth Factor-Induced Proliferation and Migration of Rat Aortic Smooth Muscle Cells

Knockdown of endogenous PDE4D expression by lentiviral vector-mediated shRNA in VSMCs. VSMCs were infected with the same amount (PDE4D-shRNA: 12.5 μL, nonsense shRNA: 6.25 μL) of lentivirus particles carrying either shRNA (titer 4 × 108 TU/mL) against PDE4D or nonsense shRNA (titer 8 × 108 TU/mL). PDE4D protein expression was determined at 48 hours postinfection. Values represent means ± SEM. E: inhibition of PDE4D expression in PDE4D shRNA-tranfected cells was calculated relative to the PDE4D expression in cells tranfected with the control nonsense shRNA. Sh-GFP: lentivirus particles carrying nonsense shRNA, Sh-PDE4D: lentivirus particles carrying shRNA against PDE4D.
