Effects of ERK1/2 blocking on LIF-induced phosphorylation of STAT3 in JEG-3 choriocarcinoma cells. (a) Representative Western blot of lysates from cells starved in serum-free medium, treated or not 2 h with the ERK1/2 blocker U0126 (10 mM), and subsequently stimulated with 10 ng/mL LIF for different time periods. The bands of all blots ( ) have been scanned for density analysis. The density of bands from phosphorylated proteins ((b): phospho-STAT3 (ser727); (c): phospho-ERK1/2; (d): phospho-STAT3 (tyr705)) has been normalized against the non-phosphorylated form. The so obtained relative density at 10 min LIF stimulation has been defined as “1” and the other values have been calculated, respectively. Bars show means, error bars show standard error. In (b) and (d) *indicates when all band densities of U0126 treated cells were compared with all without such treatment independent from LIF stimulation time (Student’s test). In (c) *indicates a significant decrease when compared with the respective value without U0126 treatment.
