Cytokine Response of Cultured Skeletal Muscle Cells Stimulated with Proinflammatory Factors Depends on Differentiation Stage

Cultured skeletal muscle cells are characterized by differential responsiveness to pro-inflammatory factors. Human myoblasts and myotubes were exposed to TNF-α (100 ng/mL) or LPS (100 ng/mL) for 24 hours. Medium containing TNF-α or LPS was removed and replaced with growth medium. Fold increase in IL-6 (white bars) and IL-1 (black bars) secretion following exposure to TNF-α or LPS was calculated to assess responsiveness of myoblasts and myotubes to stimulation with pro-inflammatory factors TNF-α or LPS. The dashed line symbolically represents the level of IL-6 or IL-1 secretion under basal conditions. Data are means  ± SEM (n = 4–6). # versus IL-6 secretion in LPS-treated myoblasts (95% CI of difference: −18.34 to −7.53) and myotubes (95% CI of difference: 2.42 to 13.23).
