Activation of MEK1/2/Nrf-2 Signaling Pathway by Epstein-Barr Virus-Latent Membrane Protein 1 Enhances Autophagy and Cisplatin Resistance in T-Cell Lymphoma

MEK1/2/Nrf-2 inhibitor suppresses the promoting effect of LMP1 on lymphoma cell resistance to DDP. The MEK1/2 specific inhibitor SL327 was added to the KHYG-1 cells overexpressing LMP1, and Nrf-2 specific activator cheirolin was added to the SNK-6 cells poorly expressing LMP1. (a) The extent of MEK1/2 phosphorylation in the KHYG-1 cells and the expression of Nrf-2 in the SNK-6 cells were detected by western blot analysis. (b) The autophagy level was observed by MDC staining. (c) The IC50 value of KHYG-1 and SNK-6 cells to DDP measured by CCK-8 assay. (d) Cell proliferation was detected by colony formation assay.
