Optimization of an In Vitro Transcription/Translation System Based on Sulfolobus solfataricus Cell Lysate

Transcriptional activity of S. solfataricus whole cell extracts. In vitro transcription reactions were performed using S. solfataricus S30 fractions with (α-32P) UTP in different experimental conditions as described in Material and Methods and Table 1. Reaction A was incubated at 60°C while reaction B was incubated at 70°C. Total RNA was extracted from the reaction mixes, and an aliquot of the samples was treated with RNase A at 37°C for 30 min. The products of in vitro transcription were subjected to nondenaturing polyacrylamide gel electrophoresis, and those incorporating (α-32P) UTP were visualized by autoradiography.
