Genotoxic and Cytotoxic Safety Evaluation of Papain (Carica papaya L.) Using In Vitro Assays

Analysis of plasmid pUC 9.1 DNA strand breaks after treatment with papain. Aliquots of pUC 9.1 plasmid DNA (200 ng) were incubated with different concentrations of papain for 40 minutes at 25 ∘ C. Each sample was mixed with loading buffer and submitted to 0.8% agarose gel electrophoresis. The assay was repeated, at least three times. Densitometric measures were obtained from gel through Gel Pro Analyzer 3.0 software. Lanes: (1) negative control (Milli-Q water); (2) positive control (H2O2 10 mM); (3) 10  πœ‡ g; (4) 25  πœ‡ g; (5) 50  πœ‡ g; (6) 100  πœ‡ g; (7) 250  πœ‡ g; (8) 500  πœ‡ g of papain.
