Cytokines and Growth Factors Stimulate Hyaluronan Production: Role of Hyaluronan in Epithelial to Mesenchymal-Like Transition in Non-Small Cell Lung Cancer

Effect of growth factors and cytokines on HA production. (a) Expression of HAS mRNA in A549 cells as visualized by ethidium bromide staining of agarose gel. A549 cells were treated with or without various growth factors and cytokines for 24 hours in RPMI-1640 supplemented with ITS following overnight serum starvation and assayed for (b) mRNA expression for HAS2 (open bar) and HAS3 (close bar) by quantitative real time RT-PCR, and (c) total HA production using DuoSet HA kit. (d) HA-dependent pericellular matrix was visualized by particle exclusion assay following 24 hours incubation with or without combination treatment with 10 ng/mL each of TGF- 𝛽 1 and IL-1 𝛽 : Panel (a), Control; Panel (b), TGF + IL; Panel (c), 2 hours incubation with 10 units/mL of Streptomyces hyaluronidase following treatment with TGF + IL.
