Development of a Serum Biomarker Assay That Differentiates Tumor-Associated MUC5AC (NPC-1C ANTIGEN) from Normal MUC5AC

Development of a surrogate MUC5AC-related antigen standard. Protein extracted from human LS174T colorectal tumor cells was prepared to generate a standard curve in the NPC-1C antigen-capture ELISA. Unlabeled NPC-1C was used to coat a 96-well microtiter plate. Following a blocking step, a detergent extract made from human LS174T colorectal tumor cells or human A549 lung tumor cells were incubated on the plates. Biotin-labeled NPC-1C was then applied to the plates to detect the bound antigen, followed by development with streptavidin-peroxidase and TMB incubation steps.
