The spectra were subtracted from pyruvate absorbance. Pyruvate concentration was evaluated by a coupled LDH assay (see Section 2). (b) Absorption spectra of 10 μM C-S lyase in the absence (solid line) and presence of 450 mM L-homoserine (dotted line) and 150 mM L-methionine (dashed line). All spectra were recorded in a solution containing 50 mM MOPS, 50 mM bicine, and 50 mM proline pH 8.5.
