p55PIK Transcriptionally Activated by MZF1 Promotes Colorectal Cancer Cell Proliferation

p55PIK expression is transcriptionally activated by MZF1 in human colon cancer cell lines SW480 and LoVo. Cells were transfected with a reporter plasmid for p55PIK promoter (−1243/+45)-p55PIK and cotransfected with or without MZF1-GFP expression plasmids or MZF1 SiRNA for 24 h. (a, b) Relative luciferase activity (RLA) was determined as described under “Materials and Methods”; (c), (d), mRNA expression of MZF1 or p55PIK was detected by real-time PCR; (e), (f) cell lysates were analyzed by MZF1, p55PIK, or GAPDH immunoblot.
