Alcohol Induced Mitochondrial Oxidative Stress and Alveolar Macrophage Dysfunction

EtOH-induced ROS production in MH-S cells. Cultured MH-S cells were either untreated (none) or EtOH treated for 1d, 3d, and 5d (0.2%, ). During the last 24 hr of EtOH treatment, some cells were cotreated with mitoT. The ROS probes CM-H2DCFDA for cellular ROS and mitoSOX for mitochondrial superoxide were added to the cell medium (30 min; 37°C). Cells were then harvested, washed, and resuspended in PBS for confocal microscopy (a) or FACS analysis ((b) and (c)). All values are expressed as mean ± SD and normalized to untreated conditions. for EtOH versus none and for 5d EtOH versus 5d of EtOH + mitoT.
