Rosiglitazone Regulates Anti-Inflammation and Growth Inhibition via PTEN

Pretreatment with PPAR antagonist did not reverse the inhibition of rosiglitazone on LPS-induced NO release and PGE2 production. RAW 264.7 cells were treated with 25 M rosiglitazone for 30 min before being stimulated with 5 g/mL LPS for 24 h. In some experiments, the cells were first treated with 10 M GW9662 for 30 min, prior to the addition of 25 M rosiglitazone for 30 min, and then followed by incubation with 5 g/mL LPS for 24 h. Cell supernatants were collected for determining NO release and PGE2 production. Data shown are representative of three individual experiments. Data are expressed as mean SEM obtained from three individual cultures. < 0.001 compared with the control, untreated group. < 0.01 and < 0.001 compared with the LPS group.
