Differential NF-κB pattern after stimulation with KMP-11 in APCs of VL patients and healthy controls. The capacity of moDCs and Ms for the activation of NF-κB was compared through intracellular staining using fluorescent conjugated anti-NF-κB antibodies on flow cytometry. Following stimulations (KMP-11 and PHA), cells were harvested and then incubated with PE-anti-p65 NF-κB antibodies, acquired and analyzed on FACS Calibur. Total amount of NF-κB was produced by M and moDC stimulated and unstimulated with rKMP-11 evaluated through FACS calibur. Immunoblotting of VL M and VL DCs (insert, Figure 5) with NF-κB antibody shows that KMP-11 triggered significant phosphorylation of a protein migrating at 65 kDa of NF-κB.
