High glucose induced inflammation, pore formation, and cell death. HRPs were firstly treated with 30 mM glucose for 12, 24, and 48 h or were treated with the indicated concentrations of high glucose (HG) or mannitol (osmotic pressure control, OP) for 48 h. (a) Cell viability was determined by MTT analysis. (b) IL-1β and IL-18 levels in the cell culture supernatant were determined by ELISA. (c) Fluorometric plots show PI uptake (RFU (%)) over time to reveal the kinetics of pore formation. (d) Cell death was monitored by measuring LDH released (LDH release (%)) by dying cells.
