Figure 2: Analysis of T-cell responses to the vaccine. (a) T-cell responses to hTERT and survivin antigens prior to (week 0) and after (week 15) vaccination were analysed in PBMCs. The cells were not incubated (−) or were incubated (+) with recombinant hTERT or survivin protein (20 μg/mL) for 6 days, and T-cell proliferation was determined by incorporation of [3H]-thymidine following overnight pulsing. The results are presented as the mean ± standard deviation of triplicate determinations. (b) Cytokine contents in culture supernatants. PBMCs were costimulated (+) or not (−) with recombinant survivin and hTERT (10 μg/mL each) for 5 days and then cytokine contents in culture supernatants were analysed by commercially available ELISA kits.
