The effect of MAPK inhibitors on FT-induced total HbF expression in K562 cells. Cells were cultured in the presence of 20 g mL−1 FT-EtOH extract with addition of ERK-inhibitor PD98059 (PD) and p38-inhibitor SB203580 (SB), respectively. (a) The number of TMB-stained cells and unstained cells was scored under inverted microscope at 200x magnification. The relative THb production of K562 cells was expressed as a percentage of number of stained cells in reference to total number of cells. (b) The concentration of HbF was determined in reference to the standard curve of HbF and expressed in percentage of HbF per total proteins. The results are expressed as mean SD of three cultures. The data are representative of three separate experiments.
