Demethoxycurcumin Retards Cell Growth and Induces Apoptosis in Human Brain Malignant Glioma GBM 8401 Cells

Apoptosis induction by DMC in GBM 8401 cells via caspase 3, 8, and 9 activations. DMC activated pro-caspase-3 degradation in GBM 8401 cells. The cells were treated with DMC (0, 12.5, 25, and 50 μM), and then (a) Western blot analysis was performed for pro- and cleaved- caspase-3. (b) Quantification of band intensities by Li-COR near-infrared imaging system. All data were reported as the means (±SEM) of at least three separate experiments. Statistical analysis used the t-test, with the significant differences determined at the level of * versus 0 μM control group. (c) The caspase 2, 3, 8, and 9 activity was analyzed by ApoAlert Caspase assay plates. The DMC induced the caspase activity of GBM 8401 cells. All data were reported as the means (±SEM) of at least three separate experiments. Statistical analysis was as cited above.
