Baicalein Triggers Mitochondria-Mediated Apoptosis and Enhances the Antileukemic Effect of Vincristine in Childhood Acute Lymphoblastic Leukemia CCRF-CEM Cells

The effects of baicalein on cell viability and apoptosis in CCRF-CEM cells.  (a) 4 × 104 CCRF-CEM cells/well were seeded in 24-well tissue culture plates and were then treated with various concentrations of baicalein for 24 h and 48 h. The cell viability was determined by the MTT assay. Each point on the graph represents the mean ± SD from triplicate wells. Similar results were obtained in three independent experiments. (b) Flow cytometry analysis of baicalein-induced apoptosis in CCRF-CEM cells. The cells were treated with the indicated concentrations of baicalein for 24 h, followed by staining with FITC-Annexin-V and PI. The data are representative of three independent experiments showing similar results. Means ± SD of the experimental replicates is presented with bar graph in the bottom. * , compared with the untreated control group. (c) Cells were treated with the indicated concentrations of baicalein for 24 h, after which the sub-G1 population was determined by propidium iodide (PI) staining and subsequent flow cytometry analysis.
