Effect of Berberine on PPARα/NO Activation in High Glucose- and Insulin-Induced Cardiomyocyte Hypertrophy

Figure 4

Concentration-dependent effects of berberine on mRNA expression of PPARα and eNOS. Cardiomyocytes were pretreated with various agents for 30 min and then stimulated by HGI (25.5  mmol/L glucose and 0.1 μmol/L insulin) for 48 h. Berberine (1, 3, or 10 μmol/L) markedly restored the decreased PPARα and eNOS mRNA expression level in a concentration-dependent manner. Fenofibrate (0.3 μmol/L) had an effect similar to berberine ( ). MK886 (0.3 μmol/L) abrogated the effects of both berberine and fenofibrate. Results are represented by mean ± SEM of 3 independent experiments. versus control; # versus HGI; versus HGI + fenofibrate (0.3 μmol/L); versus HGI + berberine (3 μmol/L). “+” or “−”: treatment with or without relevant reagent.
