Effect of doxorubicin and decursin on STAT3 signaling in multiple myeloma cells. (a–c) U266 cells were treated with decursin (80  M) and/or doxorubicin (1  M) for 24 h. Cell lysates were prepared and subjected to western blotting for (a) phospho-STAT3 and STAT3, phospho-JAK2, JAK2, phospho-Src (b) and Src, and (c) cyclin D1 and survivin. (d) U266 cells were treated with decursin (80  M) and/or doxorubicin (1  M) for 8, 16, or 24 h. Western blotting was performed for phospho-STAT3, phospho-JAK2, and cyclin D1. (e) MM.1S cells were treated with decursin (80  M) and/or doxorubicin (1  M) for 24 h. Western blotting was performed for phospho-JAK2 and cyclin D1. A representative result of three independent experiments is shown for each experiment.
