Water Extract of Dryopteris crassirhizoma Attenuates Bone Loss by Suppressing Osteoclast Differentiation and Function

Figure 1

WEDC inhibits osteoclast differentiation in bone marrow cell-osteoblast coculture. Mouse bone marrow cells and calvarial osteoblasts were cocultured in the presence or absence (negative) of VitD3 (10 nM) with vehicle (distilled water) or WEDC (10, 25, 50, and 100 μg/mL) for 6 days. (a) Representative microscopic pictures of TRAP staining are shown. (b) The number of TRAP(+)MNCs containing three or more nuclei were counted. Data are expressed as mean ± SD and are representative of three experiments. * and ** versus vehicle-treated control group. (c) Osteoblasts were preincubated with vehicle or WEDC (50 μg/mL) for 3 h and stimulated with VitD3 (10 nM) for 24 h. RANKL and OPG mRNA levels were analyzed by QPCR. Data are expressed as mean ± SD and are representative of three experiments. ** . N.S.: not significant.
