Inhibition of Adipogenesis by Oligonol through Akt-mTOR Inhibition in 3T3-L1 Adipocytes

Effect of Oligonol on the expression of adipocyte-specific transcription factors during 3T3-L1 adipocyte differentiation. 3T3-L1 preadipocytes were differentiated into adipocytes in the absence or presence of Oligonol (10, 25, or 50 μg/mL) for 8 days. (a) Total RNA was isolated from 3T3-L1 adipocytes at days 2, 4, and 8 during differentiation. mRNA expressions of PPARγ, C/EBPα, C/EBPβ, and C/EBPδ were analyzed by quantitative RT-PCR. All gene expressions were normalized using GAPDH as a reference gene. (b) Protein expression levels of PPARγ and C/EBPα were analyzed by Western blot analysis. Cell lysates were collected from 3T3-L1 cells at days 4 and 8 after induction of adipocyte differentiation.
