Inhibition of Adipogenesis by Oligonol through Akt-mTOR Inhibition in 3T3-L1 Adipocytes

Effect of Oligonol on phosphorylation of AKT, mTOR, and Foxo1 in 3T3-L1 adipocytes. 3T3-L1 preadipocytes were differentiated into adipocytes with the absence or presence of Oligonol (10, 25, or 50 μg/mL) for 8 days. (a) Oligonol decreased the serine phosphorylation of Akt in 3T3-L1 adipocytes. (b) Oligonol phosphorylated AMPK in a dose-dependent manner, and inversely attenuated the phosphorylation of mTOR-S6K in 3T3-L1 adipocytes as compared to untreated adipocytes. (c) Oligonol decreased the serine phosphorylation of FOXO1 in 3T3-L1 adipocyte. Cell lysates were collected from 3T3-L1 cells at days 4 and 8 after induction of adipocyte differentiation. Western blot analysis was performed using antibodies specific for the proteins indicated. The data shown are representative of three independent experiments.
