Aβ causes cholesterol sequestration in primary neurons. (a) Rat primary neurons (forebrain and sympathetic) cultured in serum-free medium, were treated for 24 h with 20 μM oligomeric Aβ 42 (prepared according to [66]) or with 1.5 μM U18666A, a drug extensively used to induce cholesterol sequestration. Cholesterol was examined by confocal microscopy, using filipin. (b) Neurons were treated as in (a) but fluorescent oligomeric Aβ 42 was used. Intracellular localization of cholesterol and Aβ was examined by double indirect immunofluorescence confocal microscopy using LAMP1 as a marker of late endosome/MVBs and EEA1 as a marker of early endosomes.
