CYP2E1 Sensitizes the Liver to LPS- and TNF α-Induced Toxicity via Elevated Oxidative and Nitrosative Stress and Activation of ASK-1 and JNK Mitogen-Activated Kinases

Model for the potentiation of TNFα-induced hepatotoxicity, oxidative stress mitochondrial dysfunction, and activation of MAPK by pyrazole induction of CYP2E1. Pyrazole induction of CYP2E1 coupled to TNFα induction of iNOS results in elevated oxidative/nitrosative stress in hepatocytes. This results in activation of JNK and p38 MAPK which, along with the elevated ROS/RNS, damage mitochondrial function ultimately leading to liver injury.
