CYP2E1 Sensitizes the Liver to LPS- and TNF <i>α</i>-Induced Toxicity via Elevated Oxidative and Nitrosative Stress and Activation of ASK-1 and JNK Mitogen-Activated Kinases

<table>Model for the potentiation of TNF<i>α</i>-induced hepatotoxicity, oxidative stress mitochondrial dysfunction, and activation of MAPK by pyrazole induction of CYP2E1. Pyrazole induction of CYP2E1 coupled to TNF<i>α</i> induction of iNOS results in elevated oxidative/nitrosative stress in hepatocytes. This results in activation of JNK and p38 MAPK which, along with the elevated ROS/RNS, damage mitochondrial function ultimately leading to liver injury.</table>
