(a) Secretion of IL-12p70 by DCs and DC-tumor cell hybrids in the presence of signal transduction inhibitors. DCs (black bars) or DC-tumor cell hybrids (white bars) were stimulated with LPS and supernatants were analyzed by ELISA as described in Section 2. Stimulations were performed in the absence of inhibitor (0), or in the presence of MEK inhibitor (U0126), PI3K inhibitor Wortmannin, mTOR inhibitor rapamycin, or JW74, an inhibitor of the canonical Wnt pathway. Experiments were performed in duplicate and the standard error of the mean is presented. (b) Secretion of IL-12p70 DC-tumor cell hybrids. DC-tumor cell hybrids were stimulated with LPS in the presence or absence of inhibitors as indicated. Experiments were performed in duplicate and the standard error of the mean is presented.
