Anti-IL-22 Antibody Attenuates Acute Graft-versus-Host Disease via Increasing Foxp3+ T Cell through Modulation of CD11b+ Cell Function

Figure 2

Infusion of IL-22Ab markedly suppressed engraftment of donor cells in aGVHD mice. aGVHD was induced as described in Materials and Methods. Mice were treated with PBS, anti-IgG, or IL-22Ab. After 2 weeks, mice were sacrificed, and donor and host splenic lymphocyte subsets were analyzed by flow cytometry. Splenocytes were stained with H2-Kd and H2-Kb to distinguish donor and host cells. (a) IL-22 expression in serum on day 14 for each group was tested by ELISA. (b) Survival of aGVHD mice in each group. (c) Representative plots and relative engraftment percentages and absolute numbers of donor and host cells after 14 days. (d) Representative plots of donor and host CD8+ and CD19+ cells. (e) Absolute numbers of donor CD8+ cells and host CD19+ cells shown as total numbers. Data are shown as the mean ± SEM from six independent experiments. and .
