Serum and Ectopic Endometrium from Women with Endometriosis Modulate Macrophage M1/M2 Polarization via the Smad2/Smad3 Pathway

Ectopic endometrial homogenates induced M1 to M2 polarization of macrophages. THP-1 cell-derived macrophages were subjected to LPS stimulation for 24 h, followed by treatment with either eutopic or ectopic endometrial homogenate or normal endometrial homogenate for 72 h. As detected by flow cytometric analysis (a) and RT-PCR, the percentage of CD163+ cells (b) as well as CD163 mRNA (c) and IL-10 mRNA (d) levels was significantly decreased in THP-1 cell-derived macrophages treated with ectopic endometrial homogenate, as compared with normal endometrial homogenates. However, there were no significant differences in the percentage of CD163+ cells or CD163 mRNA and IL-10 mRNA levels between THP-1 cell-derived macrophages treated with eutopic and normal endometrial homogenate.
