Blockade Effects of Anti-Interferon- (IFN-) <i>γ</i> Autoantibodies on IFN-<i>γ</i>-Regulated Antimicrobial Immunity

<div>Stimulation with anti-IFN-<i>γ</i> autoAbs decreases IFN-<i>γ</i>-induced generation of nitrite and hydrogen peroxide in differentiated macrophages. After treatment with anti-IFN-<i>γ</i> autoAb-positive patient serum (PS) or anti-IFN-<i>γ</i> autoAb-negative healthy serum (HS), (a) Western blot analysis, (b) Griess reaction, and (c) CM-H<sub>2</sub>DCFDA staining showed the biosynthesis of iNOS, the biosynthesis of NO, and the generation of hydrogen peroxide in IFN-<i>γ</i>-stimulated PMA-differentiated macrophages, respectively.
