Immobilization of DNA Aptamers on Polyester Cloth for Antigen Detection by Dot Blot Immunoenzymatic Assay (Aptablot)

Effect of 5′-tailing of aptamer. Apt 2 was synthesized with different polynucleotide ( ) tails at the 5′ end (1, untailed Apt 2; 2, 5′-poly-A-tailed; 3, 5′-poly-C-tailed; 4, 5′-poly-G-tailed; 5, 5′-poly-T-tailed) and each was spotted on polyester cloth and then UV cross-linked as before. The strips were saturated with thrombin solution followed by immunoenzymatic assay of bound thrombin as described in Methods. These experiments were repeated in a separate trial with identical results (not shown).
