Hyperphosphorylated FAK Delocalizes from Focal Adhesions to Membrane Ruffles

Pervanadate increases FAK phosphorylation at FAs. FAK/Ycam expressed U87-MG cells were treated with 100 μM pervanadate (PV) for the indicated time (minutes). Western blots probed with phospho-specific FAK Abs (against Tyr397 and Tyr861) showed time-dependent increase in phosphorylation. Endogenous FAK was detected at 125 KDa, and exogenous FAK at 200 KDa. Total amount of FAK was verified by stripping and probing with Ab against the Ct domain of FAK. Quantified numbers between blots correspond to the endogenous and exogenous phospho-FAK signal intensities normalized to total FAK. Quantifications were done using Image J software.
