Ciglitazone decreases PAI-1 expression in MCF-10A and MCF-10CA1 cells and is not reversed by PPAR-γ antagonist pretreatment. Conditioned media from cells treated with ciglitazone (0–5 μM), in the absence and presence of 5 μM GW9662, was concentrated and 10 μg total protein was separated by SDS-PAGE on a 10% polyacrylamide gel. Proteins were then transferred to PVDF membrane and probed for PAI-1 (rabbit antihuman PAI-1 antiserum) as detailed under Section 2.
