Differential Effects of Methyl-4-Phenylpyridinium Ion, Rotenone, and Paraquat on Differentiated SH-SY5Y Cells

PQ toxicity occurred in a concentration- and time-dependent manner in differentiated SH-SY5Y cells. Cells were incubated with 100, 500 and 1000 μM of PQ for 24, 48 and 72 h. (a) Increase in LDH release into the medium in differentiated SH-SY5Y cells. The delta increase in LDH release in units per liter (U/L) into the medium was calculated by subtraction of the respective controls (results were pooled from 3 different experiments, each experiment having 6 different culture wells per condition). (b) Mitochondrial dysfunction in differentiated SH-SY5Y cells evaluated by the MTT test. The viability of untreated control cells was set to 100%, and all treatments were expressed as the percentage of the respective controls (results were pooled from 3 different experiments, each experiment having 6 different culture wells per condition). The means were compared using the two-way ANOVA test, followed by the Bonferroni post hoc test (* , *** concentration versus control; ### concentration versus concentration).
