Detection of melanin content at different time points. The absorbance was measured at 475 nm and compared with the standard curve made by the serial dilution of standard melanin. The melanin content was expressed as microgram per 1 $\times$ 106 cells. Melanin content was determined in B16 cells treated with siNM_011661_001 (blue), siNM_011661_002 (pink), siNM_011661_003 (yellow), negative siRNA (green), and Lipofectamine (brown) at 24, 48, and 72 hours after transfection. Experiments were done in triplicate.
