Preferential Th1 Cytokine Profile of Phosphoantigen-Stimulated Human Vγ9Vδ2 T Cells

Phosphoantigen stimulation of γδ T cells causes no detectable increase in IL-10, IL-13, or IL-17 production. Freshly isolated PBMCs were enriched for γδ T cells using magnetic beads, and 200,000 cells were stimulated with 10 μM IPP, 10 nM HMB-PP, 50 U/mL IL-2, and/or 10 ng/mL IL-15 as indicated. The levels of IL-10 (a), IL-13 (b), and IL-17 (c) in cell supernatants were measured by ELISA after 72 hours.
