TNFα increases SHP-1 activity and protein expression. (a) TNFα (5 ng/mL) treatment of endothelial cells increased SHP-1 activity already within minutes of stimulation as assessed by pNPP-dephosphorylation assay in PAEC ( ). (b) This could also be observed in HUVEC after TNFα stimulation (5 ng/mL, 4 h) as detected by phosphorylation of SHP-1 at Y564. (c) SHP-1 protein expression was not changed within the first hours of stimulation but significantly increased after 24 hours of stimulation. *Significantly different versus baseline or as indicated at ( ).
